STAT1 (signal transducers and activators of transcription 1) is a member of the STAT family of transcription factors. STAT1 can be activated by interferon-alpha, interferon-gamma, EGF, PDGF and IL6. STAT1 is known to regulate several genes which are involved in cell growth, apoptosis, immune responses, and lipid metabolism. Further, STAT1 plays an important role in mediating cell viability in response to different cell stimuli and pathogen exposure. The STAT1 gene is located on chromosome 2. STAT1 is activated to regulate gene expression in response to extracellular signaling polypeptides including cytokines, interferons, and growth factors. After phosphorylation by JAK tyrosine kinases, STAT1 enters the nucleus to regulate transcription of many different genes. Among the seven STATs types, STAT1, STAT3, STAT5a, and STAT5b have a wide activation profile. STAT1 is activated by many different ligands including IFN family (IFN-Alpha, IFN-Beta, IFN-gamma and IL-10), gp130 family (IL-6, IL-11, LIF, CNTF, and G-CSF), and receptor tyrosine kinases (EGF, PDGF, and CSF-1). Two alternatively spliced transcript variants encoding distinct isoforms of STAT1 have been described.

STAT1 Antibody (AHO0832) in IF

Immunofluorescent analysis of STAT-1 was done on 70% confluent log phase HeLa cells. The cells were fixed with 4% paraformaldehyde for 15 minutes, permeabilized with 0.25% Triton™ X-100 for 10 minutes, and blocked with 5% BSA for 1 hour at room temperature. The cells were labeled with STAT1 Antibody (STAT1-79) (Product # AHO0832) at 1:250 dilution in1% BSA and incubated for 3 hours at room temperature and then labeled with Alexa Fluor 488 Rabbit Anti-Mouse IgG Secondary Antibody (Product # A-11059) at a dilution of 1:400 for 45 minutes at room temperature (Panel a: green). Nuclei (Panel b: blue) were stained with SlowFade® Gold Antifade Mountant with DAPI (Product # S36938). F-actin (Panel c: red) was stained with Alexa Fluor 594 Phalloidin (Product # A12381). Panel d is a merged image showing cytoplasmic and Nuclear localization. Panel e is a no primary antibody control. The images were captured at 40X magnification.

STAT1 Antibody (AHO0832) in WB

Western blot analysis of STAT1 (Fig a) was performed with 20 µg of HeLa (lane1), HeLa transfected with 0 nM, 100 nM Silencer® Select STAT1 siRNA (s277) (lane 2, 3 respectively) using Novex® NuPAGE® 10 % Bis-Tris gel (Product # NP0302BOX), XCell SureLock™ Electrophoresis System (Product # EI0002), Novex® Sharp Pre-Stained Protein Standard (LC5800). Proteins were transferred using Pierce™ G2 Fast Blotter (22834) to a nitrocellulose membrane and blocked with 5 % skim milk for 1 hour at room temperature. STAT1 was detected at 83 kDa using STAT1 Mouse Monoclonal Antibody (Product # AHO0832) at 1:500 dilution in 5 % skim milk at 4°C overnight on a rocking platform. Goat Anti-Mouse - HRP Secondary Antibody (Product # 62-6520) at 1:4000 dilution was used and chemiluminescent detection was performed using Pierce™ ECL Western Blotting Substrate (Product # 32106). (Fig b) Relative density of the bands normalized to tubulin.STAT1 Antibody (Product # AHO0832) confirms silencing of Stat1 expression (app 80%).
